ErbB2（Her2）(Human epidermal growth factor receptor 2)

Humanepidermal growth factor receptor 2 (HER2), also known as ErbB2, is a type Imembrane glycoprotein, and belongs to the epidermal growth factor (EGF)receptor family. It has no ligand binding domain of its own andtherefore cannot bind growth factors. However, it does bind tightly to otherligand-bound EGF receptor family members to form a heterodimer, stabilizingligand binding and enhancing kinase-mediated activation of downstreamsignalling pathways, such as those involving mitogen-activated protein kinaseand phosphatidylinositol-3 kinase.

It is aprotein giving higher aggressiveness in breast cancers. Approximately 30% ofbreast cancers have an amplification of the HER2 gene or overexpression of itsprotein product. Overexpression of this receptor in breast cancer is associatedwith increased disease recurrence and worse prognosis. HER2 appears to playroles in development, cancer, communication at the neuromuscular junctionandregulation of cell growth and differentiation.
