Homocysteine Methyltransferase,HMT

Homocysteine ( Hcy ) is a thiol-containing amino acid formed frommethionine during S-adenosylmethionine-dependent transmethylation reactions .It has been demonstrated that even mild or moderately elevated levels of Hcyalso increase the risk of atherosclerosis of the coronary, cerebral andperipheral arteries and cardiovascular disease. And currently the hcy level isregarded as the biomarker for cardiovascular disease diagnosis all over theworld.

Methyltransferases are a large group ofenzymes that all methylate their substrates but can be split into severalsubclasses based on their structural features. The most common class ofmethyltransferases is class I, all of which contain a Rossman fold for bindingS-Adenosyl methionine (SAM). Methyltransferases can also be grouped asdifferent types utilizing different substrates in methyl transfer reactions.These types include Protein methyltransferases, DNA methyltransferases, Naturalproduct methyltransferases, and Non-SAM dependent methyltransferases.

SAM is the classical methyl donor for homocysteinemethyltransferase (HMT , EC 2.1.1.10) , The general mechanism for methyl transfer is a SN2-like nucleophilicattack where the methionine sulfur serves as the nucleophile that transfers themethyl group to the enzyme substrate. SAM is converted to S-Adenosylhomocysteine (SAH)

Tduring this process. The breaking of theSAM-methyl bond and the formation of the substrate-methyl bond happen nearlysimultaneously. These enzymatic reactions are found in many pathways and areimplicated in genetic diseases, cancer, and metabolic diseases.
